SLU-PP-332 is a synthetic small molecule pan-agonist of the estrogen-related receptor (ERR) family, demonstrating agonist activity at ERRα, ERRβ, and ERRγ. ERRs are orphan nuclear receptors that regulate mitochondrial biogenesis, oxidative phosphorylation, fatty acid oxidation, and energy metabolism without known endogenous steroidal ligands, making them targets for chemical biology research.

Research applications include ERR transcriptional activation assays (reporter gene, FRET-based coactivator recruitment), mitochondrial biogenesis studies via PGC-1α coactivation, OXPHOS gene expression profiling in skeletal muscle and cardiac cell models, fatty acid oxidation flux analysis, and comparative selectivity studies against classical estrogen receptors (ERα, ERβ) to confirm ERR specificity.
