MOTS-C (Mitochondrial Open Reading Frame of the 12S rRNA type-C) is a 16-amino acid peptide encoded within the mitochondrial 12S rRNA gene. Its discovery identified a novel class of mitochondrial-derived peptides (MDPs) capable of nuclear translocation and gene expression modulation, positioning it as a research tool for mitochondrial-nuclear communication studies.

Mechanistic research has focused on MOTS-C’s activation of AMPK (AMP-activated protein kinase) pathways, its effects on glucose and lipid metabolism in skeletal muscle cell models, and its role in the folate-methionine cycle.
