Melanotan II is a cyclic heptapeptide analog of alpha-melanocyte stimulating hormone (α-MSH) with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2. Cyclization via lactam bridge between Asp and Lys confers enhanced proteolytic resistance and receptor selectivity relative to native α-MSH. It functions as a non-selective agonist at melanocortin receptors MC1R, MC3R, MC4R, and MC5R.

Primary research applications include MC receptor pharmacology and subtype selectivity profiling, melanogenesis pathway studies in melanocyte cultures (via MC1R/cAMP/MITF signaling), MC4R-mediated signaling in hypothalamic and brainstem cell models, and structure-activity relationship studies comparing cyclic versus linear α-MSH analogs. It is also used in competitive binding assays as a reference ligand for the melanocortin receptor family.
